Experimental Fat-Destroying Drug Could Aid in Weight Loss

Body fat could be destroyed by a drug that cuts off its blood supply, a new study suggests.

Researchers tested an experimental drug called adipotide in obese rhesus monkeys, and found the monkeys lost about 11 percent of their body weight after a 28-day treatment period and a 28-day recovery period.

The researchers essentially designed "a homing device that you can attach a payload to," said study author Renata Pasqualini, of MD Anderson Cancer Center in Houston. The target is the blood vessels that nourish fat tissue.

The researchers traced the origins of their work to the ideas of the late Dr. Judah Folkman, a prominent oncologist who suggested cancer cells would die if the blood vessels that fed them were killed. (The cancer drug Avastin came out of Folkman's research.)

"If you think about tissues you'd like to do away with, the obvious ones that come to mind are cancer cells and fat," Pasqualini told MyHealthNewsDaily.

    Lean monkeys not affected

The drug was administered by an injection, and the dose was determined by the weight of each monkey.

The animals were from a rhesus monkey population used for research where 1 to 2 percent of the monkeys are obese because of their eating patterns and a lack of interest in exercise.

In the trial, obese monkeys lost weight on the drug, while lean monkeys were not affected by taking it.

"Apparently this is something specific to white fat. We definitely don't see lean subjects losing weight," Pasqualini said.

Some concerns remain about the mechanics of the drug.

"From the perspective of a physician, when you have massive death of a tissue, the question is what happens with the debris that are released? What is the collateral damage inflicted by this mass of tissue?" said Dr. Ali Nayer, a nephrologist and clinical researcher at the University of Miami's Miller School of Medicine, who was not involved with the study.

But did the weight stay off?

While the monkeys in the study generally tolerated the drug well, Nayer said their kidney function was reduced while on the drug, which might be a source of concern, although the function returned in the recovery period.

Nayer said that because the initial studies of adipotide were conducted in mice, investigators needed to ensure the drug was working the same way in monkeys, and therefore it could be expected to work the same way in humans.

He noted that at the end of the recovery period, the monkeys were beginning to regain weight.

But given concerns about obesity, it is an important area, particularly given the lack of success and unpleasant side effects of current weight loss drugs.

"There is definitely a niche there for developing new strategies, new medications for obesity," Nayer said.

Further research is being conducted on the drug, which is licensed to the pharmaceutical company Ablaris Therapeutics, under a license from the University of Texas, with which MD Anderson is affiliated.

The study appears online today (Nov. 9) in the journal Science Translational Medicine.

im 29 years old, i went from 280lbs down to 205 then after some stressful time in my life ive gone back to 246-250 range, ive tried losing it again and been watching calorie intake and working out 3 times a week (more if i can) my BMR lists at 2300 ive been intaking on average 1500 usualy a bit less, and burning minimum 250-300 calories at the gym. I havnt lost anything sure ive gone down 5 lbs, but just weighed myself and it came right back. i dont have the time or energy to be any more commited to trying to diet/work out its hard enough as it is. Would be great if every single food item out there was 40-50% of daily allotment of sodium in every serving.oh and its been 2 months, one weigh in (i weigh once a week) i was down 5 lbs the following week i was back up 3.5lbs. Im on verge of giving up to be honest.

Hang in there. Stress does quite a number on your body.

Stop treating your weight loss like a chart- or a program you need to make x amount of progress on or you think you have failed. Focus on feeling healthy, exercising to feel better, and stop beating yourself up. You still have kept 30 pounds off. That is a success my friend. If you only loose 3 pounds a month- in 10 months you will be 30 pounds lighter. It sounds like a lame plan but you would be surprised at how effective it is at long terms weigh loss and that is what you want. There is a whole diet/exercise industry that makes you feel like you are failing at succeeding if you don't lose 8-10 pounds a month. That's crazy. Success is success. Once your body has lost a decent amount of weight (like you have) it will have a tendency to go into starvation mode too easily which actually prevents weight loss- it causes sudden and seemingly unexplained weight gain because your body thinks it needs to keep the weight because it thinks you are starving. I can't eat over 1800 calories or under 1200 or I will gain weight- I yoyo dieted too much when I was younger. I also vary when and how I eat, but the real, healthy, choices (not food labeled diet food but real food I prepare myself) get better results. It takes time- it really does. Start by getting rid of diet soda if you drink it and sugar-free/ low fat stuff, and corn syrup as well. I don't care what the argument is- my body does not like corn syrup- it can deal with straight sugar much better. But lower your sugar intake- really pay attention to food labels. Tell yourself you are going to eat 1 piece of fresh fruit or a vegatable a day and then do that. And yeah, as little fast food as possible. And you might already be doing this- if so, great. Exercise because you want to. Give yourself a couple of weeks to adjust and THEN you will start to see results. I have been where you are and there is/was no quick weight loss program that worked. This does. You will thank yourself 10+ months from now.

I wonder if there is the opposite of the medication? Something that triggers blood vessels to grow. There is less need for that but there is some need for it. People with large scars from old injuries tend to get cramps in their muscles below those old scars. It seems to be from poor circulation below the scar. Inject such a medication into the scar to open up new blood vessels and the cramps should reduce or even stop.

If this medication gets approved and ends up safe, the developers are going to be the world's first trillionaires. Far richer than any current billionaire. Millions will want to try it and I'm one of them.

Think about it, that could increase the healing process by a massive amount. It would also hurt like hell, but if a bunch of bones are broken, you're already hurting like hell. It's Skele-gro!
      • dfreybur 6 mths ago
Abhi, Thanks for the reply. My thought was about healing old scars not new ones. Such a med would have very different meaning on a new injury.

Eat healthy and exercise. It works.

      • dfreybur 6 mths ago
If that "worked" then no one would be fat. Plenty of people lose weight trying that. Nearly all of them gain it back. Clearly it does not "work" in any realistic sense.

There is a way it can "work" with this medication, if the medication ends up safe. Do the calorie arithmetic. Exercise is not sufficient to cause loss. A pound of fat is a marathon of calories but far more than a marathon of actual exercise because actual exercise does not burn 100% fat. Running 3-5 miles every other day is plenty of calories to counteract the most common regain rates, though. Exercise is sufficient to prevent regain.

A healthy diet with proper exercise does not have negative side effects... A pill might. I don't understand why humans make the easy choice over the right one.

        Because it is easy, silly.

Just leave it for the cancer people. I have struggled with obesity my whole life and this drug is not for me. I would rather continue to do it the old fashioned way.
